Good for you Greg....puts you ahead of the game in more ways than one. To see someone using their initiative like that is brilliant...and will do you no harm when it comes to making decisions about whether you want a lab based job or not. I must say I'd like to see more of that type of initiative from a school-leaver.

I seem to remember from dim amd distant undergrad days that we are in a "super-interglacial" period and that we are supposedly a few thousand years overdue for a new ice-age? I seem to remember something about ice-ages being separated by approx. 10,000 year interglacials...or am I wrong? Don't rush out and by your thermals just yet.

Err...Kettle, I'm reasonably sure that we get Brown Recluse's here in the UK......but probably not a strain that can produce that effect!!! My hair used to be reddish (faded somewhat as I grew older) and my husband is left-handed. His grandmother tried to make him right-handed. This was probably due to some old biblical crap about left-handed people being the devil's spawn (sinister and all that). I believe there is also mention of red-haired people being "bad" in the bible but I'm not sure. I hate bigotry of any sort but we all find it to some degree, Scots are mean and drunk all the time (not true - Hollywood take note), English people are all villainous and/or cowardly with a stiff upper lip (not true - Hollywood take note), Irish people are all amusing, quaint and ignorant (not true - Hollywood take note), the Welsh are all........ err.......good singers? So what I'm saying I suppose is that all so called ethnic minorities or "groups" are subjected to racism or bigotry to some degree - it will never change so live with it. My dad is a retired cop and I've lost count of the times I (and my brothers) got chased, bullied etc. by the local low-life in the small towns we lived in. Until my brothers grew very big and started beating the crap out of anyone who crossed them that is.

I've moved on a bit now....any tips on site directed mutagenesis?? I can't get it to work. I've tried changing template amounts, DMSO, used a gradient block to vary annealing temp, used a different clone, tried a kit, tried the standard lab protocol..... Any magic wands out there?

I think my husband would give a "big up" (?) to Shania Twain....he thinks she's pretty easy on the eye.

Skye makes an excellent suggestion - many principle investigators will take on summer students who want to experience real lab work. Some of them MAY even have spare funds to pay you (but not necessarily). If you're serious about science it's worth doing and you will find out the difference between undergrad practical labs and real science. And believe me, there's a huge difference.

Yes you get depressed occasionally...but that's true of all jobs. And you get the blues when stuff doesn't work...but if you enjoy problem solving and trouble shooting that's OK. There is no job on this planet that doesn't have it's down side and unless you work outdoors you will be surrounded by glass, walls etc. anyway....maybe just replace the test-tubes with computers or whatever else. I am a research scientist and I love bench science but all jobs have their down-side.
