However, I could not open the reference EE article it cites. Does anyone know if this is actually possible? which approach would be the ideal one?

Correct me if I'm wrong, but doesn't pGLO contain an amp gene as the selection marker?

I'd suggest googling some protocol for Site-Directed mutagenesis. (e.g. QuickChange method...)
