Hello,

 

For my homework (prior to work in the laboratorium) I need to find out which proteins have a natural affinity with Ni2+. I've tried googling it but unfortunately I didn't find the answer...

We'll work on the purification of a recombinant protein from E. coli, we'll be using his6 tags and some assays (including Bradford) but no chromatography :)

Histidine is an amino acid with natural affinity for nickel, so peptides with polyhistadine residues (i.e. introduced with the his tag). The wording of your question (what you mean by 'natural') is ambiguous.

Yeah, there are a lot of proteins that can bind Ni specifically or non-specifically. Running a lysate through a column usually yields you a relatively complex mix of proteins, with your target hopefully being the most abundant fraction.
