Hi Guys-

have a question , I’m trying to clone guide RNAs into a vector with Bfu1 sites. I was told to make an oligo that has

 a protospacer sequence- bfuSite/forward- reverse bfu1 site.

the protospacer is the same as the forward guide sequence without the Bfu1 site

 

i don’t understand what the protospacer is???
