oswaldonfire
Hi All, Does anybody know off the top of their head what the maximum size of DNA that can be successfully electroporated is? I have been looking all over, and have not yet found a bp size that one should not go over when trying electroporation. Is it on the order of billions, millions, or thousands of base pairs? Thank you in advance, Chris M
CharonY
It depends on what you electroporate. But as you probably know the efficiency is inversely related to the vector size. In practical terms anything above around 500 kb probably will hardly work. But you will already tend to have rather low rates above 200 kb. oswaldonfire
Thank you, CharonY. That's pretty much what I needed to know. I knew that it was inversely related, but I wasn't sure how far I could go. However, I may try a different vector to get a larger amount of DNA into the cell. oswaldonfire
Also, does anybody know if there is a limit to how much DNA can be inserted through lipofection? I am not certain, but I would think that the amount would be much higher with the liposomes than with electroporation. Thanks, Chris
