SIRSantos
I need some help about use of hydrofluoric acid to make opaque end in a coverslip, to use for 3D nuclei FISH!
Kaeroll
Hi, Perhaps if you provide some more details on what you mean, people may be able to help? I'm not entirely sure I'll be able to - but be extremely careful with HF! It's incredibly nasty stuff. Kaeroll
SIRSantos
OK! So, we use Coverslip (26x76mm) thickness 0.17 +/- 0.01 mm to grow cell in order to make FISH on 3D preserved nuclei. The problem is that we need to produce frosted ends in the coverslips. We expect that the method is simple: Immerse one end of coverslip during 30 sec in Hydrofluoric acid; rapid hash in H2O; wash slide in mixture 1:1 ethanol/ether (at least 1 h). (Solovei et al., Oxford press) The problem is we don’t see the frosted ends and if we left more time the slide brake! Any ideas?
