Genecks Posted April 22, 2010 Posted April 22, 2010 I'm looking at the action of the Sau3A enzyme. What exactly is it cutting? 5'--- What does that third hyphen represent? Picture attached.
Greippi Posted April 22, 2010 Posted April 22, 2010 (edited) It doesn't matter what the third hyphon it is. I ASSUME it could be any base and that the sequence it needs for recognition is just GATC. However, I'll just check that and get back to you in a second. Just looked it up on NEB cutter and that's correct. Edited April 22, 2010 by Greippi 1
CharonY Posted April 22, 2010 Posted April 22, 2010 The hyphens just indicate that the DNA sequences continues from there....
Mr Skeptic Posted April 22, 2010 Posted April 22, 2010 5'---GATC--- 3'---CTAG--- it gets cut as shown. The hyphens are just placeholders.
