If a protein is unfolded during the transport process then, no. Often proteins have a targetting sequence added on the end which tells the cell where it needs to go in the cell, this may interfere with the correct folding of a protein and thus make it less functional.

Even if folded correctly the protein-protein interaction with the transport system generally inhibits proper interaction with the substrate.
