apricimo Posted January 13, 2011 Posted January 13, 2011 Hello, I am working with the mentioned species and I am trying to isolate the genomic DNA. This species makes spores after some time but overall spores form throughout the growth period. Does anyone know a good strategy for isolating genomic DNA from spores? Or have experience with working with sporulating streptomyces and isolating their DNA? Thank you.
Horza2002 Posted January 15, 2011 Posted January 15, 2011 I work with S. coelicolour, however, I tend to extract the mycelia to isolate the natural product im studying. Are you attempting to isolate the DNA from the bacteria or from a spore?

Are you attempting to isolate the DNA from the bacteria or from a spore? Ideally from mycelia but it seems that spores form throughout the growth. Maybe my prep sucks. I've isolated the gemomic DNA before, but have had a hard time since. I use a bactozol kit it works nicely if there are no spores. What medium do you use for your growth to get your species to produce natural products?
Horza2002 Posted January 17, 2011 Posted January 17, 2011 I usually use LB media in a liquid state...I then place a sterlised permeable membrane over this...then add the spores onto of that. Let them grow for three days. After that I lift the membrane off which normally leaves the mycelia left which I then extract for the natural product. I don't work anything with the DNA so I don't know if theres anything wrong with your prep.
