Hello there,

I wonder if the FIX/PERM solution used during the Intra Cellular staining Could remove or kill red blood cells that could be present in the sample???

I cannot see the reason why it should, but I may be wrong. In that case, I would like to know why :D

The Permeabilisation solution that allows the antibodies to enter the cells could affect the integrity of the membrane and thus RBC may die cos of that, I was just wondering if the fixation solution could prevent this??huh.gif

Some of my colleagues in the lab noticed that after ICS experiment RBC are reduced.....

Basically all permeabilization and fixation methods put stress on cells. The composition of the solution and the resilience of the cells varies somewhat, though.

Ok, Thank yousmile.gif, Then any tips to get rid of the RBC that persist in the PBMC solution after Blood separation with Ficoll gradient???
