is this tube anaerobic or fac. anaerobic?

this is 2 pics of the same tube; i took two to see if one was better quality than the other. i tried to get as best resolution as possible.

 

The media is thioglycollate broth kept at 48 hours in 37degrees celcius. This tube was not shaken prior to taking the picture (mayb i shouldve done that?)

 

is this microorganism anaerobic or facultative anaerobic? My only concern is obviously what the red circle dictates, but i feel like the growth/settlement is too much for anaerobic

I really shouldn't even try, but judging from the pictures on the Wikipedia article I linked, I would say they're anaerobic. If my guess is accurate, a facultative anaerobic culture would be scattered in the whole tube.

But then again, I could be colossally wrong!

 

Edit: After reading a little in the article, I'm quite sure they're anaerobic. Air would hurt them, so they gather at the bottom of the tube to avoid it.

Yeah they're anaerobic. I'm assuming you incubated them aerobically? Fac anaerobes prefer oxygen, but are able to grow in anoxic environments, if you had any they'd be closer to the top. So what pwagen said was true to my knowledge, the oxygen would kill the anaerobes (or severely hamper their growth) so they grow as far down as possible.

 

P.S. From what I can gather you have a pretty decent cell count

hmmm alright; my lab partners and i thought it was anaerobic as well, but needed clarification since there was much growth from the bottom of the tube towards the middle.

 

stefan...i guess, as you said, the high cell count of my sample was what probably made it appear to accumulate so much that it created the allusion of it being facultative.

Was the medium prepared anaerobically and is a redox indicator included? The amount of cells is suspicious for only two days of pure anaerobic growth. Unless the starting titer was very high (which would not be ideal for these kind of tests).

Also, was sufficient agar in there?

The appropriate consideration would be growth on surface of appropriate agar medium.
