Hi

 

How can I using a 100bp ladder calculate the sizes of PCR products obtained with a 3' primer ?

 

 

Can someone please help me really stuck on this

Do you know what a ladder is for?

 

 

To calculate the sizes of DNA fragments ? but how by plotting a calibration graph of fragments against distance migrated ? im not sure

Yes, precisely. You have to think using semi-log scale, though (just plot it and you should see).

 

 

Okay, is it alright to use a semi-log paper with 3 cycles on the y axis and 1 cycle (upto 10) on the x axis? or does it have to be equal cycles on both x and y axis.
