Therefore, my question is if it is possible and if yes, then HOW it is possible to create a cell line with knochout of only one allele of gene? I understand general principles of generating of full knockout. Don't jugde if the queastion is too naive. I am grateful for an answer or a reference to source of wisdom.
ecoli Posted July 30, 2012 Posted July 30, 2012 off the top of my head: this isn't a complete knockout, but if the variant is different enough, you could create a miRNA to knockdown only 1 allele.
Hobble Posted August 16, 2012 Posted August 16, 2012 Are the two alleles of the gene the same (ie. homozygous) or different (heterozygous)? If the latter, then miRNA could in fact work.
