CharonY Posted October 2, 2012 Share Posted October 2, 2012 If it is a common protcol in the lab I would recommend using one of the established protocols (most protocols are not universal, almost everyone tweaks it a little bit to make it work in a given lab). You can then ask questions ideally to people who have used it, but also feel free to post questions here. If you have to create an assay from the ground up I would recommend that you check out some manufacturers of comet assay kits. They provide more standardized procedures. FLHPI Posted October 3, 2012 Share Posted October 3, 2012 Hi, It is most likely that whatever protocol you choose, you’ll need to tweak it for your laboratory set-up. I would recommend (as a starting place) the method in this paper: http://www.ncbi.nlm.nih.gov/pubmed/22844078 because it is used by a lot of the comet assay leading labs. In addition, check out www.scorecomets.com – there are web pages about ‘getting started’ with comet assay and ‘troubleshooting guide’ which might be helpful to you.
