Comet assay help

Dear

I am a student and I need to do comet assay ( that is frist time for me) and I will go to prepared buffers but I need good protocol to follow with all details not just brief .

Because I was a way for chemistry such a long time ago so I want everything explane in details plz

If it is a common protcol in the lab I would recommend using one of the established protocols (most protocols are not universal, almost everyone tweaks it a little bit to make it work in a given lab). You can then ask questions ideally to people who have used it, but also feel free to post questions here.

If you have to create an assay from the ground up I would recommend that you check out some manufacturers of comet assay kits. They provide more standardized procedures.

It is most likely that whatever protocol you choose, you’ll need to tweak it for your laboratory set-up. I would recommend (as a starting place) the method in this paper: http://www.ncbi.nlm.nih.gov/pubmed/22844078 because it is used by a lot of the comet assay leading labs.

 

In addition, check out www.scorecomets.com – there are web pages about ‘getting started’ with comet assay and ‘troubleshooting guide’ which might be helpful to you.

 

Also, the kits by Trevigen might be a good place to start.
