csiszi Posted October 8, 2012 Share Posted October 8, 2012 Hi, I am working with the good old PMSF inhibitor, that is supposed to inhibit a broad spectrum of serine proteases. But how broad is this spectrum? I am wondering how different its inhibition is to different proteases. Can one tell me if it inhibits plasmin (sounds trivial that it should so, but I could not explicitly find it) and if so with which Ki value? CharonY Posted October 8, 2012 Share Posted October 8, 2012 Well technically it will work on all serine proteases by esterification. However the issues with PMSF tends to be its stability/solubility in aqueous, high-salt and/or high pH solutions. Personally I prefer some of the newer cocktails as they give more consistent results. That being said, plasmin required relatively high amounts to be deactivated, IIRC, and together with low solubility/stability of PMSF could be an issue.
