RNase Posted September 11, 2014 Posted September 11, 2014 Is there a difference in efficiency of ligation between 5'-overhang and 3'-overhang? And do ligation products look like dsDNA? I don't understand how there are several ligation products when the PCR fragment digested by a restriction enzyme has the same sequence.
CharonY Posted September 11, 2014 Posted September 11, 2014 If you have a 5' overhang, how can you ligate without a 3'? But for multiple products there is always self-ligation, concatermerization, multiple PCR products (very common) etc.
