The skeleton of spider's silk is protein, right ?... But on testing a water sol. Of it with ninhydrin and biuret reagent no reaction is observed! How could this happen?

Ninhydrin reacts (at best) with one end of the protein chain.

If silk is a long chain (and it is) and the "end" is buried in the middle of the fibre, what would the reagent reacts with?
